Chemical composition of Kalonji:
The seeds contain numerous esters of structurally unusual unsaturated fatty acids with terpene alcohols (7%); furthermore, traces of alkaloids are found which belong to two different types: isochinoline alkaloids are represented by nigellimin and nigellimin-N-oxide, and pyrazol alkaloids include nigellidin and nigellicin. In the essential oil (avr. 0.5%, max. 1.5%), thymoquinone was identified as the main component (up to 50%) besides p-cymene (40%), a-pinene (up to 15%), dithymoquinone and thymohydroquinone. Other terpene derivatives were found only in trace amounts: Carvacrol, carvone, limonene, 4-terpineol, citronellol. Furthermore, the essential oil contains significant (10%) amounts of fatty acid ethyl esters. On storage, thymoquinone yields dithymoquinonene and higher oligocondensation products (nigellone). The seeds also contain a fatty oil rich in unsaturated fatty acids, mainly linoleic acid (50 – 60%), oleic acid (20%), eicodadienoic acid (3%) and dihomolinoleic acid (10%) which is characteristic for the genus. Saturated fatty acids (palmitic, stearic acid) amount to about 30% or less. Commercial nigella oil (“Black Seed Oil”, “Black Cumin Oil”) may also contain parts of the essential oil, mostly thymoquinone, by which it acquires an aromatic flavor.

Medicinal value of Kalonji:

  • Modern research indicates use of Kalonji oil as analgesic,anthelminticanti- bacterialanti- spasmodicanti- tumor, carminative,  diaphoretic,  galactogogue,  immunomodulatorand  laxative.
  • Anti Asthmatic: Nigella sativa oil contains nigellone, which protects guinea pigs from histamine induced bronchial spasms (explaining its use to relieve the symptoms of asthma, bronchitis, and coughing).
  • Useful in abscess and tumor: The presence of the sterol beta-sitosterol lends credence to its traditional use to treat abscesses and tumors of the abdomen, eyes, and liver.  
  • Useful for de addiction: N. sativa oil has been reported to be effective in treating opioid dependence.
  • Renal Stones: N. sativa also has been reported to reduce calculi formation in rats' kidneys.
  • Pancreatic cancer treatment: The chemical compound thymoquinone isolated from N. sativa seed oil blocks pancreatic cancer cell growth in vitro and kills the cells by enhancing the process of programmed cell death acting as an HDAC inhibitor. While the studies are in the early stages and no clinical study has started, the researchers speculated Kalonji may have some use as a preventative strategy in patients who have gone through surgery and chemotherapy or in individuals who are at a high risk of developing cancer.
  • Thymoquinone contains excellent anti-inflammatory and analgesic properties. It is also a strong antioxidant and helps cleanse the body of toxins. Both nigellone and thymoquinone work in conjunction with one another to enhance Kalonji’s action against respiratory ailments. It also provides a healthy alternative to the more commonly prescribed cortisone based therapies used by allergy sufferers. Black seed provides a rich supply of polyunsaturated fatty acids. 
  • High Blood Pressure: In a small study published in 2008, researchers found that Kalonji may help keep blood pressure in check. After eight weeks of twice-daily treatment with nigella sativa extract, patients with mild hypertension had a greater reduction in blood pressure.

Precautions:
Melanthin – on of the constituent of Kalonji is toxic in large doses and nigelline is paralytic, so this spice must be used in moderation.

Medication Interactions:
Kalonji is an antioxidant and protects cells against damage. The protective effect of Nigella sativa may interfere with the action of certain types of cancer therapies, such as radiation therapy or chemotherapy, which destroy cells that grow rapidly.
Concomitant use of Nigella sativa and antihypertensive drugs may cause a sudden drop in blood pressure levels. Low blood pressure may result in symptoms of dizziness, headache or fatigue.

Some important notes about Kalonji:

  • Kalonji is very useful as an anti infectious herb especially against round worm.
  • Kalonji is taken in painful menstrual bleeding and after delivery. It cleanses the uterus; improves lactation; and regularize the menstrual cycle.
